A typical dorsiventral leaf has distinguishable upper adaxial and lower abaxial surfaces. Both have epidermal coverings, with the upper surface commonly having a more prominent cuticle. Stomata are generally more numerous on the lower epidermis in the typical terrestrial dicot leaf; this is not an absolute rule for every habitat and species.

Mesophyll differentiates into palisade and spongy parenchyma. Palisade cells are elongated, closely arranged and usually located toward the upper side, with many chloroplasts. Spongy cells are more loosely arranged toward the lower side, leaving intercellular spaces that facilitate gas movement. Both regions can photosynthesise; spongy tissue is not simply a dead air layer.

Veins contain vascular bundles enclosed by bundle sheaths. In a typical leaf bundle, xylem lies toward the upper surface and phloem toward the lower surface. Leaf polarity therefore provides the reference for vascular orientation, unlike the centre–periphery language of a stem. In supplied counts, compare stomatal densities using equal areas or divide by area first; greater counts alone can reflect a larger observed field rather than a true difference between surfaces.

Worked example: If equal areas of upper and lower epidermis contain 12 and 36 stomata, the lower density is three times the upper. The inference depends on equal sampled areas.
